Investigadors del <\/span><a href=\"http:\/\/www.crg.eu\/\"><span style=\"font-weight: 400;\">Centre de Regulaci\u00f3 Gen\u00f2mica<\/span><\/a><span style=\"font-weight: 400;\"> (CRG) en col\u00b7laboraci\u00f3 amb cient\u00edfics del\u00a0<a href=\"https:\/\/www.upf.edu\/web\/biomed\" target=\"_blank\" rel=\"noopener\">Departament de Ci\u00e8ncies Experimentals i de la Salut, Universitat Pompeu Fabra (DCEXS-UPF)<\/a><\/span><span style=\"font-weight: 400;\">, han descobert el mecanisme pel qual les c\u00e8l\u00b7lules controlen quanta <\/span><b>mucina <\/b><span style=\"font-weight: 400;\">&#8211; el principal macrocomponent del moc &#8211; produeixen en funci\u00f3 de les seves necessitats. Aix\u00ed, les <\/span><b>c\u00e8l\u00b7lules caliciformes<\/b><span style=\"font-weight: 400;\">, les c\u00e8l\u00b7lules secretores de mucositat que es troben <\/span><b>a l\u2019epiteli de les mucoses<\/b><span style=\"font-weight: 400;\"> del sistema <\/span><b>digestiu <\/b><span style=\"font-weight: 400;\">i les vies <\/span><b>respirat\u00f2ries<\/b><span style=\"font-weight: 400;\">, poden produir grans quantitats de moc quan hi ha pres\u00e8ncia d\u2019al\u00b7l\u00e8rgens o pat\u00f2gens o b\u00e9 alliberar-ne la mida justa i de forma constant nom\u00e9s per preservar la capa mucosa. <\/span><\/p>\n<p>\u201cHi ha dos tipus de secreci\u00f3 de mucina; la basal, per mantenir la capa de moc estable, i la estimulada &#8211; per al\u00b7l\u00e8rgens o pat\u00f2gens &#8211; on es necessita una gran secreci\u00f3 de cop i volta per a protegir l\u2019epiteli. Se sabia que aquesta secreci\u00f3 est\u00e0 controlada per calci, per\u00f2 no se sabia d\u2019on venia el calci\u201d, explica\u00a0<strong>Gerard Cantero-Recasens<\/strong>, investigador post doctoral al <a href=\"https:\/\/www.crg.eu\/en\/programmes-groups\/malhotra-lab\" target=\"_blank\" rel=\"noopener\">laboratori de<strong> Vivek Malhotra<\/strong> al CRG<\/a> i primer autor dels dos articles d\u2019aquest estudi.<\/p>\n<\/div>\n<div class=\"mb10 fleft\">\n<p><span style=\"font-weight: 400;\">El mecanisme de control que els cient\u00edfics han descobert es basa en un sistema doble de <\/span><b>sensors de Calci<\/b><span style=\"font-weight: 400;\">:<\/span><\/p>\n<ol>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Dues<\/span><span style=\"font-weight: 400;\"> prote\u00efnes, el canal i\u00f2nic <strong>TRPM4 i la NCX<\/strong>,\u00a0<\/span><span style=\"font-weight: 400;\">que es troben a la superf\u00edcie de les c\u00e8l\u00b7lules i<\/span><span style=\"font-weight: 400;\"> que treballen plegades per <\/span><span style=\"font-weight: 400;\">controlar l\u2019entrada de calci a la c\u00e8l\u00b7lula.<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Un sensor de calci dins la c\u00e8l\u00b7lula, <\/span><span style=\"font-weight: 400;\">la prote\u00efna <strong>KChIP3<\/strong>, que controla la secreci\u00f3 basal de mucines per mantenir una capa \u00f2ptima a les mucoses i protegir-les. Aquest sensor actua frenant l\u2019alliberament excessiu de mucines, perqu\u00e8 en condicions normals s\u2019uneix a les ves\u00edcules secretores de mucines, impedint-ne la seva secreci\u00f3. En resposta a pics regulars de Calci intern, la KChIP3 va alliberant d\u2019una en una les ves\u00edcules secretores.\u00a0<\/span>Per\u00f2 quan TRPM4 i NCX deixen entrar molt de Calci a la c\u00e8l\u00b7lula, el sensor intern KChIP3 s\u2019uneix al Calci, deixant lliures moltes de les ves\u00edcules de cop, que aleshores poden secretar el moc. Autor: Gerard Cantero-Recasens, CRG.<\/figcaption><\/figure>\n<p><span style=\"font-weight: 400;\">Estudiant les diferents l\u00ednies cel\u00b7lulars, l\u2019equip va descobrir que el proc\u00e9s de control de la secreci\u00f3 de mucines &#8211; a trav\u00e9s de TRPM4 i NCX &#8211; \u00e9s el mateix en les vies respirat\u00f2ries i en el colon. <\/span><\/p>\n<p><span style=\"font-weight: 400;\">El grup est\u00e0 col\u00b7laborant ara amb investigadors de l\u2019<\/span><a href=\"https:\/\/www.parcdesalutmar.cat\/ca\/hospitals\/hospital-del-mar\/\"><span style=\"font-weight: 400;\">Hospital Del Mar<\/span><\/a><span style=\"font-weight: 400;\"> i l\u2019<a href=\"http:\/\/www.imim.es\/\" target=\"_blank\" rel=\"noopener\">Institut Hospital del Mar d\u2019Investigacions M\u00e8diques\u00a0(IMIM)<\/a><\/span><span style=\"font-weight: 400;\"> per identificar si \u00a0pacients amb malalties en vies respirat\u00f2ries i c\u00f2lon tenen alguna mutaci\u00f3 gen\u00e8tica en les mucines o les prote\u00efnes implicades en la seva secreci\u00f3.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Tamb\u00e9 estan treballant per trobar mol\u00e8cules qu\u00edmiques que puguin controlar aquestes vies de secreci\u00f3 de mucines. <\/span><\/p>\n<p><span style=\"font-weight: 400;\">Totes dues aproximacions, la gen\u00e8tica i la qu\u00edmica, poden esdevenir noves vies per detectar i controlar la progressi\u00f3 de <\/span><span style=\"font-weight: 400;\">malalties on es produeix massa o massa poc moc, com ara \u00a0l\u2019asma, la <strong>malaltia pulmonar obstructiva cr\u00f2nica (MPOC)<\/strong>, la <strong>malaltia de Crohn<\/strong> o el <strong>c\u00e0ncer colorectal<\/strong>.<\/span><\/p>\n<p>En aquest v\u00eddeo, el primer autor dels articles,\u00a0<strong>Gerard Cantero-Recasens<\/strong>, ens explica en primera persona com han fet aquest estudi.<\/p>\n<p>&nbsp;<\/p>\n<\/div>\n<p><iframe loading=\"lazy\" width=\"1170\" height=\"658\" src=\"https:\/\/www.youtube.com\/embed\/V6RGWtCcWVs?feature=oembed\" frameborder=\"0\" allow=\"accelerometer; autoplay; encrypted-media; gyroscope; picture-in-picture\" allowfullscreen><\/iframe><\/p>\n<p>&nbsp;<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Investigadors del CRG han identificat les prote\u00efnes que controlen la producci\u00f3 de les mucoses que protegeixen els nostres epitelis.
