{"id":46434,"date":"2025-02-07T09:35:52","date_gmt":"2025-02-07T08:35:52","guid":{"rendered":"https:\/\/ellipse.prbb.org\/?p=46434"},"modified":"2025-02-07T09:35:41","modified_gmt":"2025-02-07T08:35:41","slug":"lestructura-proteica-clau-en-levolucio-i-la-salut","status":"publish","type":"post","link":"https:\/\/ellipse.prbb.org\/ca\/lestructura-proteica-clau-en-levolucio-i-la-salut\/","title":{"rendered":"L&#8217;estructura proteica, clau en l&#8217;evoluci\u00f3 i la salut"},"content":{"rendered":"\n<p class=\"wp-block-paragraph\"><a rel=\"noreferrer noopener\" href=\"https:\/\/www.crg.eu\/en\/news\/human-domainome-reveals-root-cause-heritable-disease#2\" target=\"_blank\">Un estudi<\/a> portat a terme entre el <a rel=\"noreferrer noopener\" href=\"http:\/\/www.crg.eu\/\" target=\"_blank\">Centre de Regulaci\u00f3 Gen\u00f2mica (CRG)<\/a> i l\u2019empresa BGI de Shenzen ha determinat que <strong>les mutacions amb canvi de sentit a l\u2019ADN que causen malalties ho fan majorit\u00e0riament perqu\u00e8 <a rel=\"noreferrer noopener\" href=\"https:\/\/ellipse.prbb.org\/ca\/predir-lestabilitat-de-les-proteines-amb-mates-senzilles\/\" target=\"_blank\">desestabilitzen la prote\u00efna<\/a> que codifiquen<\/strong>. L\u2019equip ho va observar en un 60% de mutacions que van estudiar &#8211; introduint-les en c\u00e8l\u00b7lules de llevat &#8211; i es deu a que la mutaci\u00f3 resulta en un pitjor plegament de la prote\u00efna codificada.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">No obstant aix\u00f2, l\u2019equip dirigit per <strong><a href=\"https:\/\/ellipse.prbb.org\/ca\/ben-lehner-nou-membre-de-la-royal-society\/\" target=\"_blank\" rel=\"noreferrer noopener\">Ben Lehner<\/a><\/strong> tamb\u00e9 va observar <strong>v\u00e0ries mutacions que no desestabilitzen les prote\u00efnes, per\u00f2 que tamb\u00e9 causen malalties<\/strong>. Aix\u00f2 obre la porta a nous tractaments segons les conseq\u00fc\u00e8ncies de la mutaci\u00f3. Algunes de les malalties que van estudiar s\u00f3n les cataractes heredit\u00e0ries, la miopatia amb cossos reductors, el s\u00edndrome de Hay-Wells o el de Rett.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Aquest estudi ha estat possible gr\u00e0cies al <strong>projecte Dominioma Hum\u00e0 1 que ha catalogat m\u00e9s de mig mili\u00f3 de mutacions en al voltant de 500 dominis proteics humans<\/strong>. Aquestes mutacions van ser creades canviant sistem\u00e0ticament cada amino\u00e0cid per tots els altres amino\u00e0cids possibles. Tot i que \u00e9s el cat\u00e0leg d&#8217;aquest estil m\u00e9s gran fins al moment, nom\u00e9s cobreix el 2,5% de les prote\u00efnes humanes que es coneixen.<\/p>\n\n\n\n<div style=\"height:70px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<h2 class=\"wp-block-heading\"><strong>De les malalties a l\u2019arbre de la vida<\/strong><\/h2>\n\n\n\n<p class=\"wp-block-paragraph\"><a href=\"https:\/\/www.crg.eu\/en\/news\/protein-shapes-can-help-untangle-lifes-ancient-history#2\">En una altra investigaci\u00f3<\/a> del CRG, dirigida per <strong><a href=\"https:\/\/ellipse.prbb.org\/ca\/bioinformatics-for-dummies\/\" target=\"_blank\" rel=\"noreferrer noopener\">Cedric Notredame<\/a><\/strong>, <strong>es van combinar les dades sobre l\u2019estructura de les prote\u00efnes amb les de les seq\u00fc\u00e8ncies gen\u00f2miques per a determinar millor els <a href=\"https:\/\/ellipse.prbb.org\/ca\/larbre-evolutiu-mes-complet-dels-escarabats\/\" target=\"_blank\" rel=\"noreferrer noopener\">arbres filogen\u00e8tics<\/a><\/strong>. Aquests s\u00f3n clau per a entendre l\u2019evoluci\u00f3 de la vida a la terra, aix\u00ed com controlar la dels pat\u00f2gens o desenvolupar nous medicaments.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Tradicionalment, els arbres filogen\u00e8tics han estat creats nom\u00e9s amb dades de les seq\u00fc\u00e8ncies d\u2019ADN o de prote\u00efnes i mirant les semblances i les difer\u00e8ncies entre els ancestres. Per desgr\u00e0cia, els canvis entre seq\u00fc\u00e8ncies d&#8217;ADN al llarg del temps poden ser tan grans que es perd la l\u00ednia que uneix les formes ancestrals amb les modernes.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">No obstant aix\u00f2, <strong>tot i les mutacions, <a rel=\"noreferrer noopener\" href=\"https:\/\/ellipse.prbb.org\/ca\/predir-lestabilitat-de-les-proteines-amb-mates-senzilles\/\" target=\"_blank\">l\u2019estructura de les prote\u00efnes<\/a> es mant\u00e9 bastant constant<\/strong>. Aqu\u00ed, s\u2019ha combinat l\u2019estudi dels canvis de l\u2019estructura predita <strong>computacionalment<\/strong>, especialment la dist\u00e0ncia entre els amino\u00e0cids, amb els canvis de seq\u00fc\u00e8ncia d&#8217;ADN per a dibuixar m\u00e9s precisament els arbres evolutius.
