Un equip cient\u00edfic del <a rel=\"noreferrer noopener\" href=\"http:\/\/www.crg.eu\/\" target=\"_blank\">Centre de Regulaci\u00f3 Gen\u00f2mica (CRG)<\/a> va descobrir aquest fenomen en crear un atles d&#8217;esdeveniments d&#8217;empalmament durant el desenvolupament primerenc de <strong>vaques, <a href=\"https:\/\/ellipse.prbb.org\/ca\/els-animals-en-la-recerca-biomedica\/\" target=\"_blank\" rel=\"noreferrer noopener\">ratolins<\/a> i humans<\/strong>.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">L&#8217;<strong><a rel=\"noreferrer noopener\" href=\"https:\/\/ellipse.prbb.org\/ca\/lsplicing-o-empalmament\/\" target=\"_blank\">empalmament<\/a> alternatiu<\/strong> \u00e9s un mecanisme molecular que permet produir una gran diversitat d&#8217;ARNm i, per tant, prote\u00efnes, a partir d&#8217;un nombre limitat de gens. Es basa en crear diferents combinacions dels exons o seq\u00fc\u00e8ncies codificants d&#8217;un gen. A m\u00e9s, \u00e9s un <strong>mecanisme de vital import\u00e0ncia per al desenvolupament de c\u00e8l\u00b7lules complexes<\/strong> com les dels <a rel=\"noreferrer noopener\" href=\"https:\/\/ellipse.prbb.org\/ca\/identificat-un-nou-mecanisme-dautoreparacio-de-cellules-musculars-independent-de-les-cellules-mare-de-teixit\/\" target=\"_blank\">m\u00fasculs<\/a> o les <a rel=\"noreferrer noopener\" href=\"https:\/\/ellipse.prbb.org\/ca\/neurotransmissio-a-nivell-atomic\/\" target=\"_blank\">neurones<\/a>, per la qual cosa \u00e9s un proc\u00e9s altament regulat.<\/p>\n\n\n\n<div style=\"height:46px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<figure class=\"wp-block-image size-large\"><img loading=\"lazy\" decoding=\"async\" width=\"967\" height=\"600\" src=\"https:\/\/ellipse.prbb.org\/wp-content\/uploads\/2022\/05\/empalme-alternativo-CAT-967x600.jpg\" alt=\"\" class=\"wp-image-29888\" srcset=\"https:\/\/ellipse.prbb.org\/wp-content\/uploads\/2022\/05\/empalme-alternativo-CAT-967x600.jpg 967w, https:\/\/ellipse.prbb.org\/wp-content\/uploads\/2022\/05\/empalme-alternativo-CAT-361x224.jpg 361w, https:\/\/ellipse.prbb.org\/wp-content\/uploads\/2022\/05\/empalme-alternativo-CAT-768x476.jpg 768w, https:\/\/ellipse.prbb.org\/wp-content\/uploads\/2022\/05\/empalme-alternativo-CAT-1536x953.jpg 1536w, https:\/\/ellipse.prbb.org\/wp-content\/uploads\/2022\/05\/empalme-alternativo-CAT-2048x1270.jpg 2048w\" sizes=\"auto, (max-width: 967px) 100vw, 967px\" \/><\/figure>\n\n\n\n<div style=\"height:46px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<p class=\"wp-block-paragraph\">A l&#8217;<a rel=\"noreferrer noopener\" href=\"https:\/\/www.crg.eu\/en\/news\/act-sabotage-determines-mammalian-embryonic-development#2\" target=\"_blank\">estudi<\/a>, l&#8217;equip cient\u00edfic va observar que, quan els <strong>embrions nom\u00e9s comptaven amb 8 c\u00e8l\u00b7lules<\/strong>, la varietat d&#8217;ARNm alternatius era la m\u00e9s alta registrada en qualsevol c\u00e8l\u00b7lula o teixit fins a la data, mentre que, en la seg\u00fcent etapa del desenvolupament, aquesta varietat disminu\u00efa a nivells normals. Aix\u00f2 evidencia un <strong>col\u00b7lapse temporal en la regulaci\u00f3 de l&#8217;empalmament alternatiu<\/strong> <strong>durant l&#8217;activaci\u00f3 del <a rel=\"noreferrer noopener\" href=\"https:\/\/ellipse.prbb.org\/ca\/tag\/genoma\/\" target=\"_blank\">genoma<\/a> cig\u00f2tic<\/strong>; \u00e9s a dir, quan un embri\u00f3 comen\u00e7a a fer servir les seves pr\u00f2pies prote\u00efnes i ARN i deixa d&#8217;usar els materns.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Manuel Irimia<\/strong>, autor principal de l&#8217;estudi, comenta que creuen que \u00e9s un acte deliberat de sabotatge. \u201cCreiem que aix\u00f2 passa perqu\u00e8 hi ha instruccions al nostre genoma que diuen a alguns gens \u2013 en concret hem vist que s\u00f3n gens involucrats en la <a href=\"https:\/\/ellipse.prbb.org\/ca\/cancer-un-problema-de-frens-i-acceleradors\/\" target=\"_blank\" rel=\"noreferrer noopener\">reparaci\u00f3 de l&#8217;ADN<\/a> \u2013 que no facin la seva feina en aquesta etapa de desenvolupament. Creiem que en el moment que l&#8217;embri\u00f3 comen\u00e7a per primera vegada a transcriure el seu ADN poden oc\u00f3rrer molts errors que sobreactivarien aquestes prote\u00efnes. Per aix\u00f2, el mateix zigot les inhibeix abans\u201d, apunta. En efecte, s&#8217;ha vist que en aquesta etapa la <strong>resposta al dany a l&#8217;ADN \u00e9s baixa<\/strong>, i aquest &#8216;<strong>autosabotatge<\/strong>&#8216; podria ser una de les raons.<\/p>\n\n\n\n<div style=\"height:46px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\"><p>\u201cLes c\u00e8l\u00b7lules de l&#8217;embri\u00f3 fan malb\u00e9 el seu empalmament a prop\u00f2sit i ho fan per una ra\u00f3 funcional\u201d<\/p><p><strong>Manuel Irimia<\/strong><\/p><\/blockquote>\n\n\n\n<div style=\"height:46px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<p class=\"wp-block-paragraph\">Aquestes troballes resulten molt interessants des del punt de vista de la <strong>medicina regenerativa<\/strong>, ja que podria suposar un aven\u00e7 en la <strong>creaci\u00f3 de <a rel=\"noreferrer noopener\" href=\"https:\/\/ellipse.prbb.org\/ca\/cel%c2%b7lules-mare-generacio-i-regeneracio\/\" target=\"_blank\">c\u00e8l\u00b7lules totipotents<\/a> a partir de <a rel=\"noreferrer noopener\" href=\"https:\/\/ellipse.prbb.org\/ca\/de-cellules-mare-a-cellules-beta-pancreatiques-funcionals\/\" target=\"_blank\">c\u00e8l\u00b7lules mare<\/a><\/strong>. Tal com diu Irimia, \u201ccreiem que el frac\u00e0s programat de la regulaci\u00f3 de l&#8217;empalmament tamb\u00e9 passa en altres contextos fisiol\u00f2gics\u201d.
