{"id":29213,"date":"2022-04-19T12:13:37","date_gmt":"2022-04-19T10:13:37","guid":{"rendered":"https:\/\/ellipse.prbb.org\/?p=29213"},"modified":"2022-04-19T12:13:26","modified_gmt":"2022-04-19T10:13:26","slug":"una-tecnica-que-podria-revolucionar-el-desenvolupament-de-farmacs","status":"publish","type":"post","link":"https:\/\/ellipse.prbb.org\/ca\/una-tecnica-que-podria-revolucionar-el-desenvolupament-de-farmacs\/","title":{"rendered":"Una t\u00e8cnica que podria revolucionar el desenvolupament de f\u00e0rmacs"},"content":{"rendered":"\n<p class=\"wp-block-paragraph\">Aquesta imatge d&#8217;<strong>Andr\u00e9 Faure<\/strong>, investigador postdoctoral del <a rel=\"noreferrer noopener\" href=\"http:\/\/www.crg.eu\/\" target=\"_blank\">Centre de Regulaci\u00f3 Gen\u00f2mica (CRG)<\/a>, mostra l&#8217;estructura d&#8217;una prote\u00efna humana, amb els seus llocs actius marcats en groc i els llocs al\u00b7lost\u00e8rics en vermell i blau, descoberts gr\u00e0cies a una nova t\u00e8cnica.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">La majoria <a rel=\"noreferrer noopener\" href=\"https:\/\/ellipse.prbb.org\/ca\/mirar-dins-nostre-millora-el-disseny-de-farmacs\/\" target=\"_blank\">dels medicaments<\/a> estan desenvolupats per unir-se al<strong> lloc actiu <\/strong>de la prote\u00efna diana, inhibint-la o activant-la. El problema \u00e9s que aquests llocs actius solen ser molt semblants entre diferents prote\u00efnes, de manera que el medicament pot estar atacant a diverses alhora, produint efectes secundaris i reduint-ne l&#8217;efici\u00e8ncia.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Hi ha altres llocs a les prote\u00efnes, m\u00e9s amagats per\u00f2 m\u00e9s espec\u00edfics, que en unir-se a un medicament modifiquen tamb\u00e9 l&#8217;acci\u00f3 de la prote\u00efna. Aquests es diuen <strong>llocs alost\u00e8rics<\/strong>, i normalment s\u00f3n gaireb\u00e9 impossibles de detectar.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Ara un equip investigador del CRG<a href=\"https:\/\/www.crg.eu\/en\/news\/abundant-secret-doors-human-proteins-could-reshape-drug-discovery\" target=\"_blank\" rel=\"noreferrer noopener\"> ha desenvolupat una <strong>nova tecnologia<\/strong><\/a> que <strong>permet descobrir aquests llocs alost\u00e8rics &#8216;amagats&#8217; a l&#8217;estructura de la prote\u00efna<\/strong>. La t\u00e8cnica consisteix a <strong>canviar tots i cadascun dels amino\u00e0cids de la prote\u00efna<\/strong> (les peces que la formen) d&#8217;un en un, i mirar, per a cadascuna de les milers de versions de la prote\u00efna resultants, <strong>quin \u00e9s l&#8217;efecte quan s&#8217;introdueix en c\u00e8l\u00b7lules vives<\/strong> al laboratori.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">\u00ab\u00c9s un experiment de &#8216;for\u00e7a bruta&#8217; que ens permet trobar un &#8216;punt fluix&#8217; a cada prote\u00efna, un lloc on potencialment la podr\u00edem atacar amb un medicament. Fins i tot per a aquelles prote\u00efnes que fins ara es consideraven &#8216;dianes impossibles'\u00bb, explica <a href=\"https:\/\/ellipse.prbb.org\/ca\/prestigioses-beques-internacionals-i-europees-financaran-projectes-del-prbb\/\" target=\"_blank\" rel=\"noreferrer noopener\">Ben Lehner<\/a>, l\u00edder del grup.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">A m\u00e9s, segons <strong>J\u00falia Doming<\/strong>o, primera coautora de l&#8217;article, aquesta t\u00e8cnica \u00e9s \u00abm\u00e9s eficient, m\u00e9s barata i m\u00e9s r\u00e0pida\u00bb que altres que es fan servir actualment, i podria revolucionar <a href=\"https:\/\/ellipse.prbb.org\/ca\/un-acord-estrategic-per-accelerar-el-desenvolupament-de-nous-farmacs\/\" target=\"_blank\" rel=\"noreferrer noopener\">el desenvolupament de f\u00e0rmacs<\/a>.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Els i les investigadores han utilitzat la t\u00e8cnica per estudiar aquests llocs o &#8216;portes ocultes&#8217; en dos dels dominis proteics m\u00e9s comuns en humans, el SH3 i el PDZ. A m\u00e9s, han vist que aquests llocs alost\u00e8rics no tan sols poden ser encesos i apagats, sin\u00f3 modificats de formes m\u00e9s subtils, com si es tract\u00e9s del control de temperatura en un term\u00f2stat.
