{"id":28108,"date":"2022-02-23T22:39:22","date_gmt":"2022-02-23T21:39:22","guid":{"rendered":"https:\/\/ellipse.prbb.org\/?p=28108"},"modified":"2022-02-23T22:46:38","modified_gmt":"2022-02-23T21:46:38","slug":"trencar-la-barrera-que-les-cellules-de-cancer-colorectal-creen-contra-la-quimioterapia","status":"publish","type":"post","link":"https:\/\/ellipse.prbb.org\/ca\/trencar-la-barrera-que-les-cellules-de-cancer-colorectal-creen-contra-la-quimioterapia\/","title":{"rendered":"Trencar la barrera que les c\u00e8l\u00b7lules de c\u00e0ncer colorectal creen contra la quimioter\u00e0pia"},"content":{"rendered":"\n<p class=\"wp-block-paragraph\">Els laboratoris de <strong>Vivek Malhotra<\/strong> al <a rel=\"noreferrer noopener\" href=\"http:\/\/www.crg.eu\/\" target=\"_blank\">Centre de Regulaci\u00f3 Gen\u00f2mica (CRG)<\/a> i de <strong>Llu\u00eds Espinosa<\/strong> a l&#8217;<a rel=\"noreferrer noopener\" href=\"http:\/\/www.imim.es\/\" target=\"_blank\">Institut Hospital del Mar d\u2019Investigacions M\u00e8diques\u00a0(IMIM)<\/a> han col\u00b7laborat en un <a rel=\"noreferrer noopener\" href=\"https:\/\/www.crg.eu\/en\/news\/colorectal-cancers-raise-defensive-barrier-response-chemotherapy\" target=\"_blank\">estudi<\/a> que ha demostrat que els <strong>f\u00e0rmacs que es fan servir de manera habitual contra el <a rel=\"noreferrer noopener\" href=\"https:\/\/ellipse.prbb.org\/ca\/dora-romaguera-isglobal-un-dels-principals-problemes-dels-ultraprocessats-es-que-desplacen-el-consum-daliments-saludables\/\" target=\"_blank\">c\u00e0ncer colorectal <\/a>comporten un augment en la producci\u00f3 i secreci\u00f3 de mucines<\/strong>. Aquestes s\u00f3n unes prote\u00efnes que formen la mucosa, un l\u00edquid lubricant que evita la deshidrataci\u00f3 dels teixits i els protegeix davant de pat\u00f2gens i altres factors externs. <a rel=\"noreferrer noopener\" href=\"https:\/\/ellipse.prbb.org\/ca\/controlar-la-quantitat-de-mucines-per-tractar-malalties-respiratories-i-del-colon\/\" target=\"_blank\">L&#8217;augment de mucines genera una <strong>barrera f\u00edsica<\/strong><\/a> que impedeix que els f\u00e0rmacs arribin al seu objectiu, <strong>generant resist\u00e8ncia al tractament<\/strong>.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Amb la intenci\u00f3 de regular aquesta sobreproducci\u00f3 de mucina, l&#8217;equip investigador ha manipulat gen\u00e8ticament els nivells de la <strong>prote\u00efna KChIP3<\/strong>, la qual regula negativament la producci\u00f3 de mucosa &#8211; quan hi ha nivells baixos d&#8217;aquesta prote\u00efna augmenta la producci\u00f3 de mucina i viceversa. De fet, a l&#8217;estudi es va observar que <strong>les c\u00e8l\u00b7lules sense KChIP3<\/strong> (i per tant amb majors nivells de mucina) eren <strong>4 vegades m\u00e9s resistents<\/strong> als f\u00e0rmacs, mentre que aquelles que presentaven <strong>alts nivells<\/strong> d&#8217;aquesta prote\u00efna (i baixos de mucina) eren fins a <strong>10 vegades m\u00e9s sensibles<\/strong> a la quimioter\u00e0pia. Aix\u00f2 tamb\u00e9 converteix la <strong>prote\u00efna KChIP3 en un bon marcador pron\u00f2stic<\/strong> de la malaltia, ja que nivells elevats van indicar un per\u00edode de superviv\u00e8ncia lliure de malaltia m\u00e9s llarg.<\/p>\n\n\n\n<div style=\"height:60px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\"><p>Les c\u00e8l\u00b7lules sense la prote\u00efna KChIP3 eren 4 vegades m\u00e9s resistents a la quimioter\u00e0pia, mentre que aquelles que presentaven alts nivells de KChIP3 eren fins a 10 vegades m\u00e9s sensibles al tractament.<\/p><\/blockquote>\n\n\n\n<div style=\"height:60px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<h2 class=\"wp-block-heading\">Un f\u00e0rmac prometedor<\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">Tot i aix\u00f2, com diu <strong><a href=\"https:\/\/ellipse.prbb.org\/ca\/analitzant-les-criptes-del-colon\/\" target=\"_blank\" rel=\"noreferrer noopener\">Espinosa<\/a><\/strong> \u00abusar KChIP3 com a diana terap\u00e8utica t\u00e9 un inconvenient, i \u00e9s que requereix l&#8217;\u00fas de ter\u00e0pia g\u00e8nica, una tecnologia en fase de desenvolupament i molt cara d&#8217;utilitzar\u00bb. Per aix\u00f2, han investigat altres maneres de regular la producci\u00f3 de mucina, com l&#8217;\u00fas d&#8217;inhibidors qu\u00edmics.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Basant-se en estudis anteriors, l&#8217;equip investigador va provar l&#8217;<strong>SN-6, un inhibidor qu\u00edmic<\/strong> que impedeix la funci\u00f3 dels canals de sodi i calci i, per tant, <strong>bloqueja la secreci\u00f3 de mucines<\/strong>. De fet, es va veure que l&#8217;\u00fas de l&#8217;SN-6 feia que les c\u00e8l\u00b7lules de c\u00e0ncer colorectal fossin<strong> 40 vegades m\u00e9s sensibles <\/strong>als f\u00e0rmacs quimioter\u00e0pics.<\/p>\n\n\n\n<div style=\"height:60px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\"><p>L&#8217;\u00fas de l&#8217;inhibidor qu\u00edmic SN-6 va fer que les c\u00e8l\u00b7lules de c\u00e0ncer colorectal fossin fins a 40 vegades m\u00e9s sensibles als f\u00e0rmacs quimioter\u00e0pics.<\/p><\/blockquote>\n\n\n\n<div style=\"height:60px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<p class=\"wp-block-paragraph\">Aquesta troballa podria <strong>beneficiar el tractament d&#8217;altres tipus de c\u00e0ncer<\/strong>, ja que tal com diu <strong>Gerard Cantero<\/strong>, primer autor de l&#8217;estudi, \u201cla mucina es produeix a moltes parts del cos, no nom\u00e9s al tracte digestiu.
