{"id":17017,"date":"2020-07-20T11:56:15","date_gmt":"2020-07-20T09:56:15","guid":{"rendered":"https:\/\/ellipse.prbb.org\/?p=17017"},"modified":"2020-07-27T12:28:29","modified_gmt":"2020-07-27T10:28:29","slug":"facilitar-el-disseny-de-farmacs-una-plataforma-per-estudiar-les-gpcrs","status":"publish","type":"post","link":"https:\/\/ellipse.prbb.org\/ca\/facilitar-el-disseny-de-farmacs-una-plataforma-per-estudiar-les-gpcrs\/","title":{"rendered":"Facilitar el disseny de f\u00e0rmacs: una plataforma per estudiar les GPCRs"},"content":{"rendered":"\n<p class=\"wp-block-paragraph\">Al nostre cos hi ha <strong>m\u00e9s de 800 receptors acoblats a prote\u00efna G (GPCRs)<\/strong>, unes prote\u00efnes presents a la membrana cel\u00b7lular encarregades de transmetre senyals a dins la c\u00e8l\u00b7lula i involucrades en molt\u00edssims processos &#8211; i en malalties des de l\u2019Alzheimer o el P\u00e0rkinson fins al c\u00e0ncer. \u00c9s per aix\u00f2 que <strong>s\u00f3n dianes per a gaireb\u00e9 el 40% dels f\u00e0rmacs actuals<\/strong>.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Un consorci de <strong>24 laboratoris de 10 pa\u00efsos <\/strong><a rel=\"noreferrer noopener\" href=\"https:\/\/www.imim.es\/news\/325\/new-tool-for-studying-a-key-drug-target\" target=\"_blank\">acaba de fer p\u00fablica una plataforma anomenada <strong>GPCRmd<\/strong><\/a> per a que cient\u00edfics i cient\u00edfiques de diferents disciplines puguin inspeccionar i analitzar f\u00e0cilment un exhaustiu nombre de simulacions moleculars de GPCRs. Aix\u00f2 podria ajudar al disseny de nous f\u00e0rmacs amb GPCRs com a diana o a la medicina personalitzada.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">La iniciativa ha estat liderada des del <strong><a rel=\"noreferrer noopener\" href=\"http:\/\/grib.imim.es\/about-us\/\" target=\"_blank\">GRIB<\/a> &#8211; <\/strong>un programa de recerca conjunt de l&#8217;<a rel=\"noreferrer noopener\" href=\"http:\/\/www.imim.es\/\" target=\"_blank\">Institut Hospital del Mar d\u2019Investigacions M\u00e8diques&nbsp;(IMIM)<\/a> i del <a rel=\"noreferrer noopener\" href=\"https:\/\/www.upf.edu\/web\/biomed\" target=\"_blank\">Departament de Ci\u00e8ncies Experimentals i de la Salut, Universitat Pompeu Fabra (DCEXS-UPF)<\/a> &#8211;  amb important suport de l&#8217;Institut Paul Scherrer de Su\u00efssa.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><\/h2>\n\n\n\n<h2 class=\"wp-block-heading\"><\/h2>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\"><p>La plataforma <strong>GPCRmd<\/strong> permet analitzar f\u00e0cilment un exhaustiu nombre de simulacions moleculars de GPCRs. La jove cient\u00edfica \u00e9s estudiant de doctorat al Grup de recerca en desenvolupament de f\u00e0rmacs en base a GPCRs del Programa de Recerca en Inform\u00e0tica Biom\u00e8dica (GRIB), dirigit per\u00a0<strong>Jana Selent<\/strong>, investigadora Miguel Servet a l&#8217;IMIM. N\u2019hi ha de molts tipus &#8211; de fet \u00e9s la fam\u00edlia de prote\u00efnes m\u00e9s gran en humans! Per aix\u00f2 estan implicades en molts processos diferents. De fet el 40% dels f\u00e0rmacs que existeixen al mercat tenen una GPCR com a diana!<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><\/h2>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Quines dificultats comporten a l\u2019hora de desenvolupar f\u00e0rmacs contra elles?<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">S\u00f3n prote\u00efnes molt complexes. <strong>Poden causar m\u00e9s d\u2019un senyal alhora<\/strong> &#8211; un pot ser l\u2019efecte terap\u00e8utic que vols, per\u00f2 un altre pot ser un efecte secundari que no vols. I \u00e9s dif\u00edcil saber com activar la proteina per a que faci nom\u00e9s el que tu vols. <strong>La seva estructura varia segons quina mol\u00e8cula s\u2019uneix a elles<\/strong>. I quan varia la seva estructura, varia la senyalitzaci\u00f3 que envien dins la c\u00e8l\u00b7lula. Aix\u00ed, segons com s\u2019hi uneixi el potencial f\u00e0rmac, pot tenir una acci\u00f3 o una altra. Per aix\u00f2 entendre la seva estructura \u00e9s tan important, i en els darrers 10 anys s\u2019ha desxifrat l\u2019estructura 3D de moltes GPCRs.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><\/h2>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Per\u00f2 con\u00e8ixer la seva estructura est\u00e0tica en 3D no \u00e9s suficient\u2026<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Una cosa \u00e9s determinar-ne l\u2019estructura per cristal\u00b7lografia o altres t\u00e8cniques. Per\u00f2 en el cas de les GPCRs <strong>l\u2019estructura \u00e9s molt din\u00e0mica<\/strong>, i per aix\u00f2 el qu\u00e8 \u00e9s interessant \u00e9s fer despr\u00e9s <strong>simulacions amb din\u00e0mica molecular (DM)<\/strong> per explorar tot el paisatge conformacional de cada GPCR a nivell at\u00f2mic &#8212; \u00e9s a dir, mirar \u00e0tom per \u00e0tom i veure quines forces s\u2019estan aplicant els uns als altres en cada moment. Aix\u00f2 ho fem cada 2 femtosegons (0.000 000 000 000 002 segons) i anem veient com es mou la prote\u00efna &#8211; com canvia la seva estructura &#8211; \u00e0tom per \u00e0tom. Ara b\u00e9, aix\u00f2 requereix un alt nivell d\u2019expertesa aix\u00ed com molt d\u2019espai i software especialitzat&#8230;<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><\/h2>\n\n\n\n<h2 class=\"wp-block-heading\"><\/h2>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\"><p>Les simulacions amb din\u00e0mica molecular (DM) permeten explorar tot el paisatge conformacional de cada GPCR \u00e0tom per \u00e0tom, mirant quines forces s\u2019estan aplicant els uns als altres cada 2 femtosegons.<\/p><\/blockquote>\n\n\n\n<h2 class=\"wp-block-heading\"><\/h2>\n\n\n\n<h2 class=\"wp-block-heading\"><\/h2>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Per aix\u00f2 va n\u00e8ixer GPCRmd.<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">S\u00ed, com a eina per a poder <strong>compartir totes aquestes simulacions <\/strong>amb la comunitat cient\u00edfica. No nom\u00e9s entre bioinform\u00e0tics o bi\u00f3legs estructurals &#8211; que som els que fem din\u00e0mica molecular &#8211; sin\u00f3 <strong>amb investigadors o investigadores de diferents disciplines <\/strong>interessats en les GCPRs a nivell experimental: gent que est\u00e0 desenvolupant f\u00e0rmacs, que volen entendre perqu\u00e8 una determinada mutaci\u00f3 o variant en una GPCR en modifica l\u2019acci\u00f3\u2026 Per a tota aquesta gent, la informaci\u00f3 que donen les simulacions de din\u00e0mica molecular \u00e9s molt \u00fatil, per\u00f2 sovint no la fan servir perqu\u00e8 no saben d\u2019on treure-la o com interpretar-la. GPCRmd <strong>\u00e9s una plataforma oberta <\/strong>on es poden trobar aquestes simulacions de les GPCRs; de moment hi ha un 70% de totes les GPCRs de les que se\u2019n coneix l\u2019estructura, per\u00f2 esperem anar-ne afegint m\u00e9s fins arribar al 100%.
